I don't know of any study on it, but there's hardly any denying that the
stem from which the key part of the adjective derives is the stem GENES- of
the noun GENOS. Neuter nouns in stem -ES regularly show the -O- grade in
the nominative (and, of course, the accusative is identical with the
nominative. You will, of course, have checked BDAG, which cites some
literature wherein the origin and meaning of the word MONOGENHS are
disputed.
