I don't know exactly what you mean Rock.

If I am examining [vermi]compost,
1/ I take a small amount (speck/pinch) and place it on a slide, then hydrat=
e it with a drop or three of distilled water and mix & spread it across the=
slide, then look at it with the scope without a coverslip.

2/ In addition to this I mix a teaspoon or three with around ten to twenty =
times the amount of water. I use little containers with lids and shake it u=
p for 60 to 90 seconds, then leave it sit for however long with the lid off=
, examining drops from this periodically under the scope with a coverslip.

3/ In addition to this, I repeat #2 but include a food source (drop or thre=
e) like molasses, fish hydrolysate or...?

1/ tells me the current activity. 2/ tells me the current activity and the =
microbial emergent potential without additional foods
3/ tells me the microbial emergent potential with food(s)
