difference between learning to read the Septuagint and learning to read the
New Testament? What are good general reference books for both?

I have found Conybeare & Stock, Grammar of the Septuagint Greek, helpful.
They provide a large number of examples of special grammar considerations
all illustrated with examples. There is a scripture index so you can see if
the verse you are working on is listed in the text. And there are a number
of sample readings that are fully annotated to get you started.
